Characters I Currently Have
I have all the 4-star characters plus Klee, Mona, and Tartaglia.

5-Star Priorities
Venti
Disclaimer: There's no official news of a Venti re-run. This is purely hypothetical.
I now mainly want Venti because I've grown bored of the Anemo Traveler. The Anemo Traveler is a great single-target sub-DPS, but even with C4 and a shield, he's vulnerable to strong rapid-fire attacks while using his Elemental Skill, assuming it's held down. Pressing it doesn't do as much DMG.
Don't get me started with his Elemental Burst which can't suck in heavy infantry like mitachurls. I don't play Ningguang and I don't have Zhongli so I can't make his tornado stay still, on top of said heavy infantry moving a lot.
Venti's Elemental Burst has a much larger AoE. I don't know if it can suck in heavy infantry like the Anemo Mechanicus can, but still, I think it shouldn't be too hard to keep those Fatui Skirmishers in place.
Still, even light (hilichurls) and medium (large slimes, Whopperflowers) infantry annoy me. Venti is the perfect answer to these fools.
Also, lots of big numbers go brrr.
I've prepared for Venti ever since I got Mona instead of Zhongli. I got 22 Intertwined Fates, a Starbucks Venti cup, a green jacket, and Guinivere (voiced by Erika Harlacher) at the ready.
Jean
I expect to get Jean on her own Event Wish since Keqing, a non-limited character, already got hers. But even if I get Jean outside of her own Event Wish, I'll still pull for Jean because I want her at C1 for maximum pulling speed with Gale Blade.
Jean is my best waifu: she has an attractive body and clothing, a Fuck Yes to romance, and a gentle yet serious side that seeks the benefit of others.
Inflicting fall DMG on my enemies simply satisfies me, which is why I really enjoy Jean's play style. Fall DMG is unique in that it scales with the enemy's max HP and distance fallen instead of the attacker's and defender's stats, so I can use Jean right out of the box in most combat situations without too much building (and, by building, I mean steroids).
Ganyu
I bet you didn't expect this one.
I deliberately skipped the Ganyu Event Wish during 1.2 because, again, I was laser-focused on Venti. I hope they don't do a Ganyu re-run before Venti, though.
Diona and Kaeya are my most used Cryo characters mainly for spreading Cryo and preparing Melt reactions. I have Chongyun but I realized I don't enjoy his play style, so I pretty much have no Cryo main DPS.
Ganyu fills both the main DPS and sub-DPS roles well with the thicc damage her unique Aimed Shot provides and the huge AoE of her Elemental Burst. I admit the main reason I was swayed into liking Ganyu was those huge numbers.
Liyue has a lot of Geo-using enemies like Geovishaps and rock shield hilichurls, so I expect Inazuma to follow suit with Electro enemies. Cryo is super effective against Electro...and Hydro Abyss Mages.
Yes. While most Hydro enemies are weak to Electro, Hydro Abyss Mages are unique in that their shields take normal DMG from Electro but are weak to Cryo. I hate those things.
4-Star Constellations
These are much lower in priority than the 5-star characters above. I'll only pull for them once I get all the 5-star characters I want.
Xingqiu: C2-C6
All of these constellations increase Xingqiu's DMG output with his elemental abilities, making him a more valuable sub-DPS for Vaporize teams and support for Tartaglia.
Xinyan: C2-C6
Trying to get Xinyan's strongest shield is a pain because enemies like to strafe around me a lot. Even with the Anemo abilities I currently have, Sucrose sometimes doesn't hit every enemy and the Traveler knocks enemies back in multiple angles with his Palm Vortex.
C2 helps get around that pain in the butt, on top of its 100% CRIT rate allowing me to focus on stacking CRIT DMG.
For C4 and C6, more DMG is always a nice buff.
Diona: C1 and C2
Diona's Energy Cost is a painful 80. I don't plan to build her seriously since I just use her for spreading Cryo and sometimes against Geovishaps. C1 helps a little with her Energy gain.
In times when I'm stressed IRL and play too aggressively, I am heavily reliant on shields, which is why I like having more shield strength.
Noelle: All Constellations
I personally enjoy using Noelle as a main DPS. I feel Noelle does better than Ningguang in the DMG department because of her incredible attack range and her shield's Geo element.
